3,3'-Diindolylmethane (DIM) is a potent antagonist of androgen
receptor (AR) function, exhibiting a distinct structural dissimilarity from the
endogenous AR ligand dihydrotestosterone (DHT).
It is the primary digestive product of indole-3-carbinol, which is a potential
anticancer component found in cruciferous vegetables.
DIM exhibits effective anti-proliferative and anti-androgenic properties in
androgen-dependent human prostate cancer cells.
It inhibits cell proliferation in LNCaP cells and suppresses DHT-stimulated DNA
synthesis.
DIM suppresses endogenous PSA transcription and reduces DHT-induced
intracellular and secreted PSA protein levels in LNCaP cells.
It inhibits DHT-induced expression of reporter gene constructs regulated by the
prostate-specific antigen promoter in transiently transfected LNCaP cells in a
concentration-dependent manner.
